Abstrak

This research aims to determine the antibacterial activity of the crude extract and ethanol fraction of kirinyuh leaves (Chromolaena odorata) against pathogenic bacteria using the column chromatography method. The test bacteria used are Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The research was conducted at the Microbiology Laboratory and the Chemistry Laboratory of UNIMED. The extraction method was performed by maceration using 96% ethanol as the solvent. The crude ethanol extract of kirinyuh leaves in this study used concentrations of 25%, 50%, and 75%. Subsequently, the fractionation of the kirinyuh leaf ethanol extract was carried out using column chromatography with a chloroform:ethyl acetate (7:4) eluent. The antibacterial activity test results showed that the crude ethanol extract of kirinyuh leaves had a greater potential to inhibit the growth of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa compared to the antibacterial activity test of the ethanol fraction. Fractionation of the column chromatography results with F1 showed weak antibacterial potential against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa bacteria, with inhibition zones of 3.05 mm, 3.8 mm, and 2.65 mm, respectively. Treatment F2 exhibited moderate potential in inhibiting the growth of Escherichia coli and Staphylococcus aureus bacteria, with inhibition zones of 5.5 mm and 6.1 mm, while showing weak potential against Pseudomonas aeruginosa with an inhibition zone of 4.65 mm. F3 did not show potential in inhibiting the growth of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa bacteria.
