Abstrak

Kirinyuh is a weed commonly found in Indonesia and is often considered harmful by the community. This plant contains many metabolite compounds that act as antibacterials, so the endophytic bacteria from kirinyuh leaves are assumed to produce antibacterial compounds similar to those of their host. The research was conducted to isolate and characterize endophytic bacteria from kirinyuh leaves, as well as to observe their antibacterial activity against Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922, and Salmonella typhi ATCC 14028. Isolation was carried out using the spread plate method, and antibacterial activity tests were conducted on Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922, and Salmonella typhi ATCC 14028 bacteria. The research results show that 6 endophytic bacterial isolates were obtained from kirinyuh leaves, including 3 Gram-negative rod-shaped endophytic bacterial isolates, namely isolates BEDK 1, BEDK 5, and BEDK 6. There are 2 Gram-Positive rod-shaped isolates, namely BEDK 2 and BEDK 4, and there is 1 Gram-Negative round-shaped isolate, namely BEDK 3. Antibacterial activity testing showed that there are 3 isolates capable of inhibiting the test bacteria Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922, and Salmonella typhi ATCC 14028. Isolate BEDK 6 is capable of inhibiting Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922. The antibacterial activity of isolate BEDK 4 shows weak antibacterial properties. The research results show that the highest inhibition zone was then identified using BLAST analysis and phylogenetic tree analysis using the MEGA 11 application. The identification results show that isolate BEDK 2 has the closest phylogenetic relationship of 99.93% with Bacillus cereus strain YR1 16S ribosomal RNA gene, partial sequence.
